You gain S class on the levels by reaching a set number of points. Points are gained by painting things (and repainting things), completing challenges, picking up color atoms, getting gold medals and finding the hidden purple swirl collectibles throughout the levels. Completion time is not a factor. If you dont get the S class the first time you play a level, you can re-select it from the map screen and try again. Note that doing so resets the entire level (except for lightbulbs and pictures). I have my scores, prior to the bonuses, listed at the end of each level's collectible guide. You can use that to gauge if you will receive the "S" class prior to ending the level.
