It opens up after you do the first tomb and get to the area where you can assemble the statue. The challenge tomb is on the right on the way to the next tomb, the game will point it out to you.
Just run through it , stepping on the four pressure plates and upen the gate at the end. The trick is to not step on a plate with a skull twice.
