Doctor (Lyle - Boston, during the farmers' third side-mission)
Farmers (Warren & Prudence - Frontier)
Huntress (Myriam - Homestead)
Inn-keepers (Oliver & Corrine - Homestead)
Lumberers (Godfrey & Terry - Homestead/story-related)
Miner (Norris - Boston)
Tailor (Ellen - New York)
Woodworker (Lance - Homestead)
Priest (Father Thomas - Homestead)

You are only required to complete each characters initial mission denoted by the Homestead Mission icon on the map. You do not need to finish each of the characters individual side quests for this trophy.
