You will need

Winrar
The ROM (Games) Files
Tutorial for getting/using DeSmuME
Getting
1)Go to the link above and chose your operating bit
2)Download and open it with winrar
3)Extract to a specified folder or your desktop (I recommend a folder)
Using
1)Open up DesmuMe
2)In the top left corner click on file, open, and find wherever you saved your ROM
3)Click on the ROM you want to play then game should load up and enjoy playing!
Other Information
How to use you Xbox 360 Controller (Wired)
1)In the top left corner you should see Config
2)Under Config you should see Controller Config Second from the top
3)Click on Controller Config then click on UP and use your controller's stick to move up and it should go down the list of controls and you can set it up to whatever you like
4)Click OK when finished and you can now use your 360 controller
How to find your operating bit
1)Go to the start menu
2)Control Panel
3)Find System and it should tell you from there

Katsumi If anyone is looking to play Pokemon on their PC i highly recommend pokemon sapphire. It was my first and i found it really enjoyable. I never knew that you could use a wired Xbox 360 controller with the emulator though, i will be sure to try it out in the near future
I think you just have to configure the remapping of the buttons and your all good that why it's not common most will just use pc controls
