Hold LT to aim at the enemy player then when your aim is on the player press RT to shoot the gun then heypresto you have one hit killed a player.

Go for head shots as that is the area that sustains the most damage per shot by any weapon in any game that I know of. Aiming a little above the head will help as well since it is a shotgun. Also, resist the temptation to hip fire it unless you are at point blank range. I hope that this will assist you.

This is my setup.
Remmy with a Stock, Long barrel, and quick draw.
Everything else you can choose. But this one is good for me at least.
