Step 1: Get the Biolab Laptop Dongle from the Scientist in the church (simply track opportunity 'Absolution'). Let her confess in the church and then wait for her in the morgue. Pick up the dongle after knocking her out.
Step 2: Get into the mansion area over the rooftops. Make your way through the kitchen and into the observatory (beware of security cameras).
Step 3: Get VHS tape from the observatory and put it in the player downstairs to lure Silvio (simply track opportunity 'Memory Lane'). When he arrives, wait for him to send the guards away. Then take him out and hide his body in a crate.
Step 4: Go upstairs and over the rooftops to reach Francesca, your target person. While she is in the room with the laptop kill her when the guard walks to the other side of the room. Put her body in the crate behind the laptop.
Step 5: Backtrack to the observatory and go downstairs into the lab. There's a laptop on the roof of a building at the start of the lab. Activate it with the Biolab Laptop Dongle (that you got in step 1) to destroy the virus immediately. Now rush to the sea plane to exit the area. Don't take too long or the area gets crowded with guards.
You are also allowed to make manual saves in between and reload them in case something goes wrong.
Also keep in mind you must hide all bodies, not get spotted and only kill the targets (you can subdue / knock out other people but have to hide their bodies).
Completing this challenge gives you a ton of points towards Sapienza Mastery Level 20. Reaching level 20 unlocks the "Amalfi Pearl" trophy / achievement.
