Destroy the boss of the second area
The boss of the second area appears immediately after killing all of the enemies in level L6. As soon as the last one is dead, you'll see three exclamation marks pop up on the bottom of the screen. This is where the boss is going to enter from, so move out of the way. Damaging this boss entails shooting him right in the little tail where he shoots from, meaning you'll need to get right in the line of fire to hit him.
